TIMELESS siRNA refers to small interfering RNA molecules designed to silence the expression of the *TIMELESS* (TIM) gene. The TIMELESS protein is a highly conserved component of the circadian clock and plays a pivotal role in maintaining genome stability by coordinating DNA replication with the cell cycle and responding to replication stress. In various malignancies, including breast, lung, and colorectal cancers, TIMELESS is frequently overexpressed, where it promotes tumor cell proliferation, survival, and resistance to chemotherapy. By leveraging the RNA interference (RNAi) mechanism, TIMELESS siRNA induces the sequence-specific degradation of TIM mRNA, leading to reduced protein levels. While primarily utilized as a research-use-only (RUO) tool to investigate chronobiology and DNA damage repair pathways, it is also studied in preclinical models as a potential therapeutic strategy to sensitize cancer cells to DNA-damaging agents and inhibit tumor progression.
