A TIMP-1 neutralizing antibody is a therapeutic agent designed to bind and neutralize Tissue Inhibitor of Metalloproteinases-1 (TIMP-1). TIMP-1 is a multifunctional glycoprotein that canonically acts as an endogenous inhibitor of matrix metalloproteinases (MMPs), but also functions independently as a cytokine-like signaling molecule to promote cell survival, proliferation, and epithelial-mesenchymal transition (EMT). In oncology, TIMP-1 is often upregulated in the tumor microenvironment and is associated with drug resistance, immunosuppression, and metastasis, including leptomeningeal and brain metastasis. Neutralizing antibodies targeting TIMP-1 are being investigated in preclinical models to block its interaction with cell surface receptors (such as CD63) and restore matrix metalloproteinase activity or alleviate local immunosuppression, thereby reducing tumor burden and metastasis.
