TINK-101 is an investigational allogeneic, off-the-shelf chimeric antigen receptor-modified invariant natural killer T (CAR-iNKT) cell therapy targeting CD7-positive malignancies. Developed by Tinkeso Therapeutics, it is designed to treat T-cell acute lymphoblastic leukemia (T-ALL), T-cell lymphoma, and acute myeloid leukemia (AML). The therapy utilizes a specific subset of iNKT cells that naturally lack CD7 expression (approximately 44% of human iNKT cells), which allows for the expression of a CD7-targeting CAR without the need for genome editing to prevent fratricide (self-killing). TINK-101 leverages a proprietary expansion platform capable of producing over 100 doses from a single donor leukopak. Preclinical studies have demonstrated significant efficacy, including doubled survival time in T-ALL mouse models and long-term cancer-free survival in AML models.
