TIP-CAR T cell is an investigational CD19-targeted Chimeric Antigen Receptor (CAR) T cell therapy designed to treat B-cell malignancies. It incorporates a 37-amino-acid segment (TIP) from the *Herpesvirus saimiri* tyrosine-protein kinase-interacting protein into the CAR-BB-CD3ζ domain. This TIP motif enhances the recruitment of the T cell kinase LCK to the immune synapse, thereby strengthening proximal signaling and improving T cell activation kinetics. This innovative design aims to overcome limitations of current CAR T therapies, such as relapse, T-cell exhaustion, and limited persistence, by promoting durable antitumor immunity and memory formation. Preclinical studies have shown enhanced target-cell killing, rapid and sustained tumor clearance, and prolonged survival in xenograft models.
