Tipepidine hibenzate is a synthetic, non-opioid antitussive and expectorant of the thiambutene class. Developed in Japan in 1959, it is primarily used to suppress cough and facilitate sputum discharge, especially for symptoms associated with cold and bronchitis. Its mechanism of action involves inhibition of G protein-coupled inwardly rectifying potassium channels (GIRK), which modulates monoaminergic neurotransmission in the brain. This action not only underlies its antitussive effect but also leads to increased dopamine levels in the nucleus accumbens, contributing to potential antidepressant-like effects. Tipepidine has been investigated for use beyond cough suppression, including attention-deficit hyperactivity disorder (ADHD), depression, and obsessive-compulsive disorder[2][3][5]. It is administered orally and has an established safety profile in both children and adults.
