Overview

Tiqudosin is a small molecule non-opioid analgesic developed by Nanning Maple Leaf Pharmaceutical for the treatment of moderate to severe cancer-related pain. Chemically, it is 12-chlorotetrahydropalmatine, a chlorinated derivative of tetrahydropalmatine belonging to the tetrahydroprotoberberine (THPB) alkaloid family. Tiqudosin primarily functions as a dopamine D2 receptor antagonist, a mechanism that provides analgesia through pathways distinct from traditional opioid narcotics. This non-opioid approach is intended to offer effective pain relief while potentially reducing the risk of common opioid-related adverse effects such as respiratory depression, constipation, and physical dependence. In clinical development, tiqudosin has been evaluated as an injectable formulation, often used as an adjunct to enhance the efficacy of standard opioid therapies like morphine. As of 2021, the drug has advanced to Phase III clinical trials in China for the management of cancer pain.
