Overview

TIR-C is a topical oligonucleotide-based immunomodulatory treatment being developed by TIRmed Pharma for the treatment of atopic dermatitis (eczema). The active ingredient, TIR-01, is a chemically stabilized, single-stranded, non-coding, non-complementary oligonucleotide. It acts as an immunomodulator by selectively inhibiting endosomal Toll-like receptors (TLRs) and preventing the uptake of pro-inflammatory agents into endosomal compartments. This mechanism modulates immune responses directly at the site of inflammation in the skin, inducing regulatory T cells (iTregs) to restore immune balance and reduce inflammation. Preclinical studies have demonstrated that a short course of treatment—five topical applications over two weeks—can provide sustained symptom relief for up to 15 months.
