tirabrutinib + entospletinib

tirabrutinib + entospletinib is a fixed combination regimen of two small molecule kinase inhibitors developed for the treatment of B-cell malignancies, notably chronic lymphocytic leukemia (CLL). Tirabrutinib is a selective inhibitor of Bruton tyrosine kinase (BTK), interfering with B-cell receptor (BCR) signaling to suppress malignant B-cell proliferation and survival. Entospletinib is a selective inhibitor of spleen tyrosine kinase (SYK), another critical mediator of BCR signaling. Together, they target distinct, complementary kinase pathways to deepen therapeutic response and potentially overcome resistance seen with monotherapy. Combination therapy has demonstrated acceptable safety and preliminary efficacy in studies of relapsed/refractory CLL and other B-cell lymphomas[1][3][4][5][6][7].
