Overview

Tiragolumab + rituximab is an experimental combination therapy for cancer, specifically tested in relapsed or refractory non-Hodgkin lymphoma. Tiragolumab is a fully human IgG1 monoclonal antibody that targets TIGIT, an immune checkpoint receptor on T cells and natural killer (NK) cells. It blocks TIGIT's interaction with CD155 and CD112 (poliovirus receptor family), reversing immune suppression and restoring anti-tumor T cell and NK cell activity. Rituximab is a chimeric monoclonal antibody targeting CD20, a cell surface antigen on pre-B and mature B lymphocytes; this induces B cell lysis mainly through complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, and induction of apoptosis. The combination leverages both direct B cell cytotoxicity from rituximab and tumor immune microenvironment modulation by tiragolumab, aiming to improve outcomes in B cell lymphomas. This combination is in clinical development and currently not approved for any indication.
