Tiratricol is a synthetic thyroid hormone analogue and a metabolite of the natural thyroid hormones T3 (triiodothyronine) and T4 (thyroxine). It acts as an agonist at thyroid hormone receptors, mimicking the effects of endogenous thyroid hormones. Tiratricol is indicated for the management of thyroid hormone resistance syndrome and has been used as adjuvant therapy in thyroid cancer. It has also been investigated for use in Allan-Herndon-Dudley syndrome (MCT8 deficiency), hyperlipidemia, localized lipodystrophy, obesity, and reduction of corticosteroid-induced atrophy. Tiratricol exhibits organ-selective thyromimetic effects with augmented hepatic and skeletal actions compared to levothyroxine[1][5][6][8].
