Tiropramide is an antispasmodic small molecule drug used primarily to treat spastic pain and muscle spasms associated with gastrointestinal, hepatobiliary, urinary, and gynecological disorders such as irritable bowel syndrome and biliary dyskinesia[1][2][3][4][5]. Its mechanism of action involves increasing intracellular cyclic adenosine monophosphate (cAMP) concentrations in smooth muscle cells, likely by inhibiting cAMP catabolism through phosphodiesterase inhibition. This leads to relaxation of smooth muscle. Additionally, tiropramide enhances calcium binding to the sarcoplasmic reticulum in smooth muscle cells, preventing calcium from interacting with contractile proteins and further promoting muscle relaxation[1][3][6]. Tiropramide is a derivative of tyrosine/phenylalanine[2], marketed worldwide for these indications.
