Overview

Tislelizumab is a humanized IgG4 monoclonal antibody that targets programmed death receptor-1 (PD-1), functioning as an immune checkpoint inhibitor. By binding to PD-1 on T lymphocytes, it blocks the interaction with its ligands (PD-L1 and PD-L2), thereby reactivating T cells and enhancing anti-tumor immune responses. The antibody was engineered to minimize binding to Fcγ receptors on macrophages, reducing antibody-dependent phagocytosis of T cells and potentially improving anti-tumor efficacy. Developed by BeiGene, tislelizumab is approved for multiple cancer indications including classical Hodgkin lymphoma, urothelial carcinoma, non-small cell lung cancer, hepatocellular carcinoma in China; esophageal squamous cell carcinoma in the EU and US; and HER2-negative gastric or gastroesophageal junction adenocarcinoma in combination with chemotherapy[3][4][5][6][7].
