This triplet combination therapy consists of tislelizumab, a humanized IgG4 anti-PD-1 monoclonal antibody; golidocitinib, a potent and selective Janus kinase 1 (JAK1) inhibitor; and selinexor, a first-in-class selective inhibitor of nuclear export (SINE) targeting XPO1. The regimen is currently being evaluated in a Phase Ib/II clinical trial sponsored by Fudan University for the treatment of relapsed or refractory extranodal natural killer/T-cell lymphoma (R/R ENKTL). By simultaneously blocking the PD-1/PD-L1 immune checkpoint, inhibiting the dysregulated JAK/STAT signaling pathway, and restoring the nuclear localization of tumor suppressor proteins via XPO1 inhibition, this combination aims to provide a synergistic therapeutic effect in patients who have progressed after standard L-asparaginase-containing chemotherapy.
