Overview

This is a **combination regimen** of three drugs: - **tislelizumab**: a monoclonal antibody immunotherapy that acts as a programmed death receptor 1 (PD-1) inhibitor, blocking the interaction between PD-1 on T cells and PD-L1 or PD-L2 on tumor cells, thus enhancing antitumor immune response. - **5-fluorouracil**: a fluoropyrimidine small molecule chemotherapy drug that inhibits thymidylate synthase, impeding DNA synthesis and leading to cytotoxicity in rapidly dividing cells. - **cisplatin**: a platinum-based chemotherapy agent that causes DNA crosslinking and apoptosis in cancer cells. This combination is **approved as first-line treatment** for adults with unresectable or metastatic HER2-negative gastric or gastroesophageal junction adenocarcinoma with PD-L1 expression, and is used in clinical trials for advanced esophageal squamous cell carcinoma. The regimen provides immunotherapeutic activity alongside traditional cytotoxic chemotherapy[1][2][4].
