Drug intelligence / Profile preview
Tislelizumab + afatinib is an investigational combination regimen consisting of tislelizumab, a humanized IgG4 monoclonal antibody targeting the programmed death-1 (PD-1) receptor, and afatinib, an oral, irreversible ErbB family tyrosine kinase inhibitor that targets EGFR (ErbB1), HER2 (ErbB2), and HER4 (ErbB4). Tislelizumab acts as an immune checkpoint inhibitor, preventing PD-1 from interacting with its ligands (PD-L1 and PD-L2) to restore T-cell-mediated antitumor immune responses. Afatinib blocks ErbB receptor signaling pathways that drive tumor cell proliferation and survival, while also inducing immunogenic cell death to release tumor antigens. This combination is designed to achieve synergistic antitumor activity by combining targeted molecular inhibition with immune checkpoint blockade. It has been evaluated in the Phase 2 neoCHANCE-1 trial as a neoadjuvant treatment for patients with locally advanced head and neck squamous cell carcinoma (HNSCC) and locally advanced hypopharyngeal squamous cell carcinoma (HPSCC).
