This multimodal therapeutic regimen combines the immune checkpoint inhibitor tislelizumab, the multi-target receptor tyrosine kinase (RTK) inhibitor anlotinib, and ET-SPACE near-infrared whole-body hyperthermia. Tislelizumab is a humanized IgG4 monoclonal antibody that targets PD-1, preventing its interaction with PD-L1 and PD-L2 to restore anti-tumor immune responses. Anlotinib inhibits multiple RTKs, including VEGFR, FGFR, PDGFR, and c-Kit, exerting anti-angiogenic and anti-proliferative effects. The ET-SPACE device provides whole-body thermal stimulation (reaching rectal temperatures of 38.5–39 °C), which is intended to sensitize the tumor to the pharmacological agents and enhance the overall anti-tumor immune response. This combination is being evaluated in a Phase 2 study led by Pengyuan Liu for the treatment of metastatic castration-resistant prostate cancer (mCRPC).
