Overview

Tislelizumab + BCG is a combination therapy under investigation for high-risk non-muscle-invasive bladder cancer (HR-NMIBC). Tislelizumab is a humanized IgG4 monoclonal antibody that targets programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1 and PD-L2, thereby restoring T-cell function and enhancing immune-mediated antitumor responses. Bacillus Calmette–Guérin (BCG) is an intravesical immunotherapy using live attenuated Mycobacterium bovis to stimulate local immune responses in the bladder. The combination aims to leverage both systemic checkpoint inhibition and local immune activation for improved efficacy in HR-NMIBC, particularly in patients unresponsive to or relapsing after standard BCG therapy[2][4][7].
