This combination neoadjuvant regimen consists of tislelizumab, carboplatin, and albumin-bound paclitaxel (nab-paclitaxel). Tislelizumab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated anti-tumor immune responses. Carboplatin is a platinum-based alkylating agent that induces DNA cross-linking, thereby inhibiting DNA synthesis and causing cell death. Albumin-bound paclitaxel is a microtubule inhibitor that stabilizes microtubules and prevents their disassembly, leading to cell cycle arrest and apoptosis. This specific chemo-immunotherapy doublet (TP) plus checkpoint inhibitor regimen is being investigated by Sun Yat-Sen Memorial Hospital of Sun Yat-Sen University as a neoadjuvant treatment for patients with early-stage oral squamous cell carcinoma (cT1-2N0M0) to enhance pathological response and improve event-free survival prior to surgical resection.
