This combination therapy regimen consists of the PD-1 checkpoint inhibitor tislelizumab, the innate immune activator Imprime PGG, and the standard-of-care chemotherapy agents cisplatin and gemcitabine. Developed by HiberCell (following its acquisition of Biothera Pharmaceuticals) in collaboration with BeiGene, the regimen was primarily investigated for the treatment of advanced or metastatic urothelial carcinoma. Imprime PGG is a yeast-derived beta-1,3/1,6 glucan that acts as a pathogen-associated molecular pattern (PAMP), priming innate immune cells such as neutrophils and monocytes to enhance their anti-tumor activity when combined with tumor-targeting antibodies or checkpoint inhibitors. Tislelizumab is a humanized IgG4 monoclonal antibody designed to minimize binding to FcγR on macrophages, potentially avoiding antibody-dependent cellular phagocytosis (ADCP) of T cells. The addition of chemotherapy aims to induce immunogenic cell death and provide synergistic anti-tumor effects by leveraging both the adaptive and innate immune systems.
