Overview

Tislelizumab + decitabine is a combination therapy being evaluated for the treatment of relapsed or refractory classic Hodgkin lymphoma (cHL). Tislelizumab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor. It is specifically engineered to minimize binding to Fcγ receptors on macrophages, thereby preventing antibody-dependent cellular phagocytosis (ADCP), a mechanism that can limit the efficacy of anti-PD-1 antibodies. Decitabine is a hypomethylating agent that inhibits DNA methyltransferase, leading to the reactivation of epigenetically silenced genes. In the context of classic Hodgkin lymphoma, decitabine is hypothesized to enhance the immunogenicity of tumor cells and sensitize them to PD-1 blockade. This regimen is being investigated by the Institute of Hematology & Blood Diseases Hospital in China as a second-line salvage therapy, often followed by autologous stem cell transplantation (ASCT) consolidation.
