Overview

This combination therapy consists of tislelizumab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, administered alongside the GX chemotherapy regimen, which includes gemcitabine and capecitabine. Tislelizumab functions as a checkpoint inhibitor, enhancing the immune system's ability to detect and destroy tumor cells by blocking the PD-1/PD-L1 pathway. Gemcitabine is a nucleoside analog that interferes with DNA synthesis by inhibiting ribonucleoside reductase and competing for incorporation into DNA, while capecitabine is an oral prodrug of 5-fluorouracil that inhibits thymidylate synthase. This specific combination is being evaluated by Fudan University as a first-line treatment for recurrent or metastatic nasopharyngeal carcinoma (NPC), comparing its efficacy and safety profile against the standard-of-care tislelizumab plus gemcitabine and cisplatin (GP) regimen.
