Overview

This combination therapy consists of the PD-1 inhibitor tislelizumab, the anthracycline liposomal doxorubicin (PLD), and the alkylating agent ifosfamide. It is being investigated as a first-line treatment for specific subtypes of unresectable or metastatic soft tissue sarcoma. Tislelizumab is a humanized monoclonal antibody that blocks the interaction between PD-1 and its ligands, enhancing T-cell anti-tumor activity. Liposomal doxorubicin provides a sustained release of doxorubicin, which intercalates into DNA and inhibits topoisomerase II, while ifosfamide causes DNA cross-linking. The combination aims to leverage the immunogenic cell death induced by chemotherapy to enhance the efficacy of the checkpoint inhibitor.
