This drug combination is a multi-modality therapeutic regimen consisting of **tislelizumab**, **LM-302**, and **S-1**. Tislelizumab is a humanized IgG4 monoclonal antibody designed to bind to the Programmed Cell Death Protein 1 (PD-1) receptor, thereby inhibiting the PD-1/PD-L1 pathway and enhancing the body's anti-tumor immune response. LM-302 (also known as TPX-4589) is an antibody-drug conjugate (ADC) that targets Claudin 18.2 (CLDN18.2), a protein frequently overexpressed in gastric and gastroesophageal junction cancers; it delivers a cytotoxic payload (monomethyl auristatin E, MMAE) directly to the tumor cells. S-1 is an oral fluoropyrimidine chemotherapy agent composed of tegafur (a 5-FU prodrug), gimeracil, and oteracil. This combination is primarily being investigated as a neoadjuvant treatment for patients with Claudin 18.2-positive locally advanced gastric or gastroesophageal junction adenocarcinoma to improve pathological complete response rates.
