Tislelizumab + oxaliplatin + capecitabine is a combination regimen used primarily for the treatment of advanced or metastatic gastric and gastroesophageal junction adenocarcinoma, as well as other gastrointestinal cancers. Tislelizumab is a monoclonal antibody that targets programmed cell death protein 1 receptor (PD-1), blocking its interaction with PD-L1 and PD-L2 to enhance anti-tumor immune responses. Oxaliplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking, leading to apoptosis in rapidly dividing cells. Capecitabine is an oral prodrug of 5-fluorouracil, which inhibits thymidylate synthase and disrupts DNA synthesis in cancer cells. The combination leverages both immunotherapy and cytotoxic chemotherapy mechanisms to improve clinical outcomes in patients with HER2-negative, unresectable or metastatic disease[1][5][8].
