Tislelizumab + oxaliplatin + S-1 is an investigational perioperative and neoadjuvant chemoimmunotherapy regimen for locally advanced gastric cancer and gastroesophageal junction adenocarcinoma. Tislelizumab is a humanized anti-programmed cell death protein 1 monoclonal antibody that restores antitumor T-cell activity by blocking PD-1 checkpoint signaling. Oxaliplatin is a platinum chemotherapy agent that forms cytotoxic DNA crosslinks. S-1 is an oral fluoropyrimidine fixed-dose combination of tegafur, gimeracil, and oteracil; tegafur is converted to 5-fluorouracil, gimeracil inhibits dihydropyrimidine dehydrogenase to increase 5-fluorouracil exposure, and oteracil reduces gastrointestinal activation of 5-fluorouracil. The regimen is commonly referred to as tislelizumab plus SOX, where SOX denotes S-1 plus oxaliplatin.
