Overview

A multi-modality *anticancer regimen* consisting of the immune checkpoint inhibitor **tislelizumab** (a PD-1–targeted monoclonal antibody), **paclitaxel** (a microtubule-stabilizing cytotoxic agent), **cisplatin** (a platinum-based DNA-damaging agent), and **radiotherapy** (ionizing radiation, such as intensity-modulated radiotherapy or low-dose radiotherapy). This combination is under investigation or early clinical use for locally advanced or unresectable cancers, notably head and neck squamous cell carcinoma (HNSCC) and esophageal squamous cell carcinoma (ESCC). - Tislelizumab acts by blocking PD-1 on T cells, preventing cancer-mediated immunosuppression. - Paclitaxel and cisplatin provide synergistic cytotoxic effects via disruption of cell division and induction of DNA damage. - Radiotherapy induces local tumor cell death and may enhance anti-tumor immune responses. Neoadjuvant and induction regimens with this combination have shown promising pathological complete response rates and manageable safety in early-phase trials[1][4][6].
