Drug intelligence / Profile preview
Tislelizumab + pemetrexed disodium + carboplatin is a combination regimen consisting of the anti-PD-1 monoclonal antibody tislelizumab, the antifolate chemotherapy agent pemetrexed disodium, and the platinum-based alkylating chemotherapy agent carboplatin. This triplet regimen is used as a first-line treatment for advanced or metastatic non-squamous non-small cell lung cancer (NSCLC) without actionable genomic alterations (such as EGFR or ALK mutations). Tislelizumab works by binding to the PD-1 receptor on T-cells, blocking its interaction with PD-L1 and PD-L2, thereby restoring anti-tumor T-cell activity. Pemetrexed disodium inhibits key folate-dependent enzymes required for nucleotide synthesis, while carboplatin binds to DNA to form interstrand and intrastrand crosslinks, inhibiting DNA replication and transcription. Together, the combination provides synergistic anti-tumor activity by combining immune checkpoint inhibition with cytotoxic chemotherapy.
