Drug intelligence / Profile preview
The combination of tislelizumab and platinum-etoposide is a chemoimmunotherapy regimen primarily developed for the first-line treatment of extensive-stage small cell lung cancer (ES-SCLC). Tislelizumab is a humanized IgG4 monoclonal antibody designed to bind with high affinity to the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell-mediated anti-tumor activity. It is uniquely engineered to minimize binding to Fcγ receptors on macrophages, preventing antibody-dependent cellular phagocytosis of T cells. Platinum-etoposide is a standard chemotherapy backbone consisting of a platinum agent (either carboplatin or cisplatin) and etoposide, which induce DNA crosslinking and topoisomerase II inhibition, respectively, leading to tumor cell apoptosis. In the phase 3 RATIONALE-312 trial, adding tislelizumab to platinum-etoposide significantly improved overall survival (OS) and progression-free survival (PFS) compared to chemotherapy alone.
