Overview

Tisotumab vedotin is an antibody-drug conjugate (ADC) used to treat recurrent or metastatic cervical cancer. It consists of a human monoclonal antibody targeting tissue factor (TF) conjugated via a protease-cleavable linker to monomethyl auristatin E (MMAE), a potent microtubule-disrupting agent. The drug binds to TF, which is overexpressed on many solid tumors, leading to internalization and release of MMAE inside the cancer cell, causing cell cycle arrest and apoptosis. Tisotumab vedotin may also block TF’s interaction with factor VIIa, potentially inhibiting angiogenesis and tumor proliferation, and can induce bystander killing of neighboring cells. It was developed as a targeted therapy for cancers expressing high levels of tissue factor.
