Tisporin is a clear ophthalmic solution formulation of cyclosporine (0.05%) developed by Hanlim Pharm. It is specifically indicated for the treatment of moderate to severe dry eye disease (keratoconjunctivitis sicca). Unlike the standard oil-in-water emulsion formulations such as Restasis, Tisporin is designed as a transparent aqueous solution, which is intended to improve patient comfort, reduce the incidence of blurred vision upon administration, and ensure uniform dosing. The active ingredient, cyclosporine, is a potent immunosuppressive agent that acts by binding to cyclophilin. This complex then inhibits calcineurin, a calcium-calmodulin-dependent phosphatase. This inhibition prevents the dephosphorylation and nuclear translocation of the nuclear factor of activated T-cells (NFAT), thereby blocking the transcription of pro-inflammatory cytokines like interleukin-2 (IL-2). By suppressing T-cell mediated inflammation on the ocular surface, Tisporin helps to increase tear production and improve the health of the corneal and conjunctival epithelia.
