Drug intelligence / Profile preview
Tivunatamig (PF-08634404, SSGJ-707) is an investigational, fully humanized bispecific tetravalent immunoglobulin G4 (IgG4) monoclonal antibody that simultaneously targets programmed cell death protein 1 (PD-1) and vascular endothelial growth factor A (VEGF-A). Originally developed by 3SBio and its subsidiaries (including Sunshine Guojian and Shenyang Sunshine Pharmaceutical) using its proprietary CLF2 platform, the asset was licensed globally (excluding China, with an option for China) to Pfizer in 2025. Tivunatamig is designed to combine immune checkpoint inhibition and anti-angiogenesis in a single agent, enhancing anti-tumor activity by localizing anti-VEGF activity and reversing VEGF-mediated immunosuppression in the tumor microenvironment. It is currently in Phase 2 and Phase 3 clinical development for various solid tumors, including non-small cell lung cancer (NSCLC), metastatic colorectal cancer (mCRC), advanced/recurrent endometrial cancer, and urothelial cancer.
