TJ-L1A3 is an investigational bispecific monoclonal antibody (BsAb) designed to target both programmed cell death 1 ligand 1 (PD-L1) and lymphocyte activation gene 3 (LAG-3). Originally developed by I-Mab Biopharma and subsequently transitioned to TJ Biopharma, the therapeutic is engineered to enhance antitumor immune responses by simultaneously blocking two key immune checkpoint pathways that tumors exploit to evade immune surveillance. By targeting both PD-L1 and LAG-3, TJ-L1A3 aims to overcome resistance to conventional anti-PD-(L)1 monotherapies and promote synergistic T-cell activation within the tumor microenvironment. The candidate is currently in Phase 1 clinical development for the treatment of multiple cancers.
