TJ-L1IF is a novel antibody-cytokine fusion protein, specifically a PD-L1/IFN-α fusion molecule designed for the treatment of PD-1/PD-L1 resistant tumors. The molecule is a biological product comprising an anti-PD-L1 antibody fused to interferon alpha (IFN-α), with the aim of delivering the cytokine directly to the tumor microenvironment, thereby enhancing anti-tumor immune responses while limiting systemic toxicity. The IFN-α2b domain in TJ-L1IF is masked with a polyethylene glycol (PEG) group through a protease-cleavable linker, rendering the cytokine inactive in circulation and selectively activated in the tumor microenvironment where tumor-associated proteases cleave the linker. This prodrug strategy enhances the safety and efficacy profile, making TJ-L1IF a differentiated approach for solid tumors that are resistant to conventional immune checkpoint inhibitors. The drug is being developed by I-Mab[7][9][10].
