Overview

TJ-RO4B is a novel tri-specific antibody designed for cancer therapy. It employs a bi-paratopic design to target two distinct epitopes of Receptor tyrosine kinase-like orphan receptor 1 (ROR1) and also targets the CRD3/4 domain of 4-1BB (CD137). This unique design aims to achieve ROR1-dependent conditional activation of 4-1BB, thereby enhancing T cell stimulation specifically within the tumor microenvironment. The drug is engineered as a Y-body-like antibody with an intact human IgG1 Fc region, which enables potent antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP) against ROR1-positive cells and contributes to anti-tumor activity and depletion of regulatory T cells (Tregs). TJ-RO4B is currently in development as a promising candidate for treating ROR1-positive malignancies, demonstrating a favorable safety profile and typical monoclonal antibody-like pharmacokinetics in preclinical studies.
