TJ073 is a GalNAc-conjugated small interfering RNA (siRNA) developed by Tuojie Biotech for the treatment of obesity and metabolic-associated steatohepatitis (MASH), formerly known as non-alcoholic steatohepatitis (NASH). The drug utilizes the RNA interference (RNAi) pathway to specifically target and degrade the mRNA of **Hydroxysteroid 17-Beta Dehydrogenase 13** (HSD17B13) in hepatocytes. HSD17B13 is a lipid droplet-associated protein primarily expressed in the liver, and genetic variants resulting in its loss of function have been associated with protection against chronic liver diseases and metabolic dysfunction. By silencing HSD17B13, TJ073 aims to modulate lipid metabolism and provide therapeutic benefits for weight management and liver health. The asset is currently in Phase 1 clinical development, where its safety, tolerability, and pharmacokinetics are being evaluated in overweight or obese subjects.
