TJ106 is a preclinical-stage biparatopic, dual-payload antibody-drug conjugate (ADC) being developed by Phrontline Biopharma for the treatment of HER2-expressing breast cancer. The drug utilizes a bispecific antibody approach to target two distinct extracellular domains of the Human Epidermal Growth Factor Receptor 2 (HER2), which is intended to enhance tumor selectivity and receptor internalization. TJ106 is uniquely conjugated with two different cytotoxic payloads: a topoisomerase I inhibitor and a microtubule inhibitor. This dual-payload strategy aims to deliver two independent mechanisms of cell death simultaneously to the target cell, potentially overcoming drug resistance mechanisms associated with single-payload ADCs and extending the treatment response in pan-HER2 breast cancer populations.
