Overview

TJH2201 is a novel, bivalent anti-CD47 monoclonal antibody engineered to provide potent tumor suppression while minimizing hematologic toxicity, specifically red blood cell (RBC) destruction. Developed by Innovent Biologics in collaboration with Tongji Hospital, TJH2201 targets the CD47/SIRPα "do not eat me" axis, an immune checkpoint that tumor cells exploit to evade phagocytosis by macrophages. Unlike earlier CD47 antagonists such as Hu5F9, which often cause severe anemia due to RBC agglutination, TJH2201 is designed to avoid this toxicity through a SIRPα-independent mechanism while maintaining high-affinity binding and robust pro-phagocytic activity. Preclinical studies in xenograft models of B-cell lymphoma (Raji) and acute myeloid leukemia (MV-4-11) have demonstrated that TJH2201 effectively suppresses tumor growth without inducing significant weight loss or hematologic safety concerns.
