Overview

TK-4202 is an in vivo T-cell receptor engineered T-cell (TCR-T) therapy being developed by T-knife for the treatment of PRAME-expressing solid tumors. The therapy is designed to target the tumor-associated antigen PRAME (preferentially expressed antigen in melanoma). TK-4202 is engineered with synthetic biology enhancements, specifically incorporating a FAS-based checkpoint converter and costimulatory armoring. These modifications are designed to overcome the immunosuppressive tumor microenvironment, enhancing T-cell fitness, persistence, and survival. TK-4202 is currently in the discovery/preclinical stage of development.
