Overview

TK-6302 is a next-generation, "supercharged" T cell receptor engineered T cell therapy (TCR-T) developed by T-Knife Therapeutics. It is designed to target PRAME (Preferentially Expressed Antigen in Melanoma), an antigen highly expressed across a range of solid tumors, including cancers of the skin, uterus, ovaries, lungs, breasts, esophagus, kidneys, cervix, and head & neck. The therapy incorporates several proprietary enhancements to improve T cell fitness and persistence within the immunosuppressive tumor microenvironment and to increase durability of response. These enhancements include a high-affinity PRAME-targeting TCR (T cell receptor), an activating chimeric CD8 co-receptor that can engage CD4+ T cells for co-stimulation upon antigen recognition, and a FAS-based checkpoint converter that boosts peripheral fitness and prevents apoptosis in the tumor environment. Preclinical data demonstrate superior anti-tumor efficacy compared to other clinical-stage PRAME-targeted approaches[1][4][5][6][8].
