Overview

TK216 + vincristine is an investigational combination therapy under clinical evaluation for the treatment of relapsed or refractory Ewing sarcoma and related tumors. TK216 is a first-in-class small molecule that targets the EWS-FLI1 fusion protein, a key oncogenic driver in Ewing sarcoma, by disrupting its interaction with RNA helicase A (RHA), thereby inhibiting downstream transcriptional activity essential for tumor growth[1][4][5]. Vincristine is a well-established vinca alkaloid chemotherapy agent that inhibits microtubule formation, leading to mitotic arrest and cell death[2][6][8]. Preclinical studies have shown that combining TK216 with vincristine enhances anti-tumor effects compared to either agent alone[1][3]. The combination is currently being studied in Phase II clinical trials for patients with relapsed or refractory Ewing sarcoma[4].
