TKB662 is an experimental small molecule immunosuppressive agent that was developed by Isotechnika, a Canadian biopharmaceutical company (now part of Aurinia Pharmaceuticals). It belongs to a class of compounds designed to inhibit calcineurin, a protein phosphatase essential for T-cell activation. TKB662 was primarily investigated for the prevention of organ transplant rejection and the treatment of autoimmune diseases. The drug was engineered to provide a more predictable therapeutic response and a superior safety profile compared to first-generation calcineurin inhibitors like cyclosporine. A key clinical goal for TKB662 was to reduce the requirement for high-dose corticosteroids in transplant recipients, thereby minimizing steroid-associated side effects such as hypertension and glucose intolerance. Development of TKB662 was ultimately discontinued as the company prioritized its lead candidate, voclosporin.
