TKM-100802 is an investigational antiviral drug developed for the treatment of Ebola virus disease (EVD). It is a lipid nanoparticle formulation containing small interfering RNAs (siRNAs) that target two essential Ebola virus gene products: L polymerase (Lpol), which is involved in viral transcription and replication, and Viral Protein 35 (VP35), which suppresses the host immune response. By silencing these genes, TKM-100802 aims to inhibit viral replication and modulate the immune response to infection. The drug was prioritized by the World Health Organization as a leading experimental therapy during the West Africa Ebola outbreak. Clinical evaluation included preclinical studies in non-human primates and phase 1 trials in humans. However, its development was limited by safety concerns such as cytokine release syndrome, leading to partial clinical hold status with regulatory authorities. Efficacy data are limited due to compassionate use settings where patients also received other therapies[3][4][5].
