Overview

TKM-ALDH2 is an investigational RNA interference (RNAi) therapeutic developed to inhibit the expression of mitochondrial aldehyde dehydrogenase 2 (ALDH2), an enzyme involved in the metabolism of acetaldehyde, primarily to address alcohol dependence and alcohol use disorders. By silencing ALDH2, TKM-ALDH2 increases acetaldehyde levels following alcohol intake, thereby deterring alcohol consumption through aversive physiological responses. The drug was designed as a novel alternative to existing therapies, such as disulfiram, aiming for improved efficacy and tolerability. Primary indications under investigation include alcohol dependence and alcohol use disorder[9].
