TKN9 is a novel aza-artemisinin derivative being investigated for its anti-fibrotic properties, specifically for the treatment of progressive pulmonary fibrosis (PPF) and interstitial lung diseases (ILD). Developed by researchers at the University of Tokyo, TKN9 functions by inhibiting the TGF-β1-driven differentiation of human lung fibroblasts into pathogenic myofibroblasts. In preclinical studies using primary human lung fibroblasts derived from both non-fibrotic and end-stage ILD lungs, TKN9 demonstrated a concentration-dependent suppression of key myofibroblast and extracellular matrix markers, including alpha-smooth muscle actin (ACTA2) and Collagen Type I (COL1A1). This mechanism positions TKN9 as a potential therapeutic agent to halt or reverse the progressive fibrosis characteristic of various lung diseases.
