TL-021 is a half-life extended bispecific antibody (HLE BsAb) currently in the discovery phase of development by TrueLab Biopharmaceutical. It is designed to simultaneously target Tumor Necrosis Factor-like Ligand 1A (TL1A) and the α4β7 integrin, two validated pathways in the pathogenesis of inflammatory bowel disease (IBD). TL1A is a cytokine that promotes mucosal inflammation and tissue fibrosis, while α4β7 integrin is responsible for the selective trafficking of pro-inflammatory lymphocytes to the gastrointestinal tract. By inhibiting both targets, TL-021 aims to provide a synergistic therapeutic effect for patients with Ulcerative Colitis (UC) and Crohn's Disease (CD) who may not respond adequately to existing monotherapies.
