TL117 is a small molecule inhibitor of phosphoinositide 3-kinase (PI3K), specifically targeting the alpha isoform (PI3Kα), developed by Suzhou Junde Biotechnology. It is currently in clinical development for the treatment of relapsed or metastatic squamous cell carcinoma of the head and neck (SCCHN). The PI3K/AKT/mTOR signaling pathway is frequently dysregulated in various malignancies, including head and neck cancers, where it promotes tumor cell proliferation, survival, and resistance to therapy. TL117 is designed to selectively inhibit PI3K activity, thereby disrupting downstream oncogenic signaling. It is being evaluated in clinical trials both as a monotherapy and in combination with chemotherapy agents such as paclitaxel to improve outcomes in patients with advanced head and neck squamous cell carcinoma.
