TL117 is a phosphatidylinositol 3-kinase (PI3K) inhibitor, while paclitaxel is a well-established microtubule-stabilizing antineoplastic agent. The combination is designed to leverage the tumor-suppressive effects of PI3K pathway inhibition and the cytotoxic effect of microtubule stabilization to maximize anticancer activity[1]. Paclitaxel works by binding to β-tubulin and stabilizing microtubules, inhibiting their disassembly, resulting in cell cycle arrest and apoptosis[4][5].
