Aro Biotherapeutics is developing a Centyrin-based therapeutic targeting Tumor Necrosis Factor-like Ligand 1A (TL1A) for the treatment of immune and inflammatory diseases. This program utilizes Aro's proprietary Centyrin platform, which consists of small, stable, engineered proteins derived from the third fibronectin domain of human tenascin-C. Centyrins are designed to combine the high affinity and specificity of monoclonal antibodies with the stability and small size of small molecules. In this specific program, the Centyrin platform is employed to enable the targeted delivery of drug payloads, such as oligonucleotides, to specific tissues or cell types to modulate the TL1A pathway. TL1A is a clinically validated pro-inflammatory cytokine and a member of the TNF superfamily that plays a key role in mucosal inflammation and fibrotic signaling, making it a significant target for conditions like inflammatory bowel disease.
