TLC-1235 is an investigational, oral, controlled-release mitochondrial protonophore being developed by OrsoBio to increase whole-body energy expenditure and improve systemic metabolic health in severe metabolic disorders. By partially uncoupling mitochondrial oxidative phosphorylation, based on a controlled-release formulation of the classical protonophore 2,4-dinitrophenol (DNP), TLC-1235 is designed to raise energy expenditure in a sustained, lower-peak manner to enhance insulin sensitivity and favorably remodel lipid metabolism while mitigating historical safety issues associated with non–controlled-release DNP.[5][3] Preclinical studies in rodent and nonhuman primate models of NAFLD/NASH, type 2 diabetes, and lipodystrophies have shown consistent improvements in insulin resistance, NASH, dyslipidemia, weight, and broader cardiovascular risk markers, and the compound is currently in IND-enabling development with NIDDK SBIR funding for eventual clinical evaluation in patients with severe lipodystrophies and other severe metabolic disorders.[5][1][12][9]
