Overview

TLC-3595 is a potent, selective, orally bioavailable small molecule inhibitor of acetyl-CoA carboxylase 2 (ACC2). It acts as an allosteric inhibitor and is designed to increase fatty acid oxidation (FAO), reduce ectopic lipid accumulation in tissues such as skeletal muscle and liver, and improve insulin sensitivity. The drug represents a novel therapeutic approach for type 2 diabetes by targeting the underlying metabolic dysfunction associated with insulin resistance. TLC-3595 has completed Phase 2a clinical trials in patients with insulin resistance/type 2 diabetes and is being developed for further studies. The compound may also have potential applications in other conditions characterized by impaired FAO, including cirrhosis with myosteatosis-associated sarcopenia and non-alcoholic steatohepatitis (NASH). Originally developed by Shionogi, TLC-3595 was acquired by OrsoBio for continued development[1][2][3][6][8].
